It's one of the best understood techniques and a sensitive method for collagen. The method is most frequently used on paraffin sections of tissues fixed adequately (at least 24 hours but ideally 1 or 2 weeks) in a neutral buffered formaldehyde solution. The collagen will be stained red with pale yellow background, and nuclei will be stained blue-black. This protocol has not been tested on frozen sections.
