The unique design of the TMA-Cassettes allows users to build 3 sized (2, 3, 5 mm) tissue microarrays easily and quickly. No need to make paraffin recipient blocks therefore save time and effort and avoid common problem of donor block cores not matching recipient block holes. Another advantage of using the TMA-Cassettes is that users do not need to fill up all the cassette holes if not needed (you will need to fill up all holes in regular paraffin recipient blocks even they are not needed). Just leave the cassette holes empty and they will be automatically filled up with liquid paraffin during the embedding step. So the users can custom make their own TMA blocks as desired. The disposable TMA-Cassettes can be reused if needed so it is ideal for beginners to practice the TMA techniques before valuable tissues are used.

Procedure

  1. Compare the reference slide (i.e, H&E stained slide) of Donor Block on microscope stage for position marking with an oil pen.
  2. Extract tissue samples that are marked with an oil pen from Donor Blocks using Tissue Microarray Tip (F1).
  3. Deliver extracted tissue sample cores to TMA Cassette holes (F2).
  4. Repeat steps 1-3 until the cassette is completed. If less holes are needed, just leave the cassette holes empty and they will be filled up with liquid paraffin during embedding step.
  5. Apply one drop of water glue to base mold and continue to rub and spread until it becomes sticky (about 30 times) (F4). Note: Base molds should be cleaned with boiling water and dried to make the water glue spread evenly. The stickiness (glue characteristic) lasts 3 to 5 minutes after applied.
  6. Put TMA Cassette into base mold with cutting side face down and push the tissue cores to the bottom with stick (F5).
  7. Embedding: pour liquid paraffin on TMA cassette and all the empty spaces will be filled with liquid paraffin (F6).
  8. Incubate the TMA cassette along with the base mold in 60°C oven for 60 minutes.
  9. Remove the TMA cassette from oven and solidify on cold plate (F7).
  10. Cutting as routine paraffin sectioning (F8).
  11. Staining using desired methods (F9).

*** Base molds should be cleaned with boiling water and dried each time after using them.

*** When you need more TMA sections after cutting the block to the end (to the edge of cassette), put the cassette in the oven for 5 minutes at 60°C and repeat steps 5-10.
