Chemicon NeuN staining- high background!
« on: December 06, 2006, 04:28:37 PM »
Hi,

I've been trying to do a Chemicon NeuN stain (1:100) on paraffin basal ganglia and have been using proteinase K (50ug/ml) for 5 min digestion.  The citrate bath just seems to damage my tissue badly.  I've been getting really high background staining when using the DAB chromagen and no signal with the SG chromagen from VectorLabs.  Please help!

I am confused. Did you use both Proteinase K and Citrate buffer for antigen retrieval?
