Hi -- Does anyone have any experience staining for the HA tag in vivo? I infected murine lung with adenovirus expressing an HA-tagged transgene. I sac'd the mice on day 4 after infection and prep'd for frozen section. I fixed in cold acetone and stained with Covance (formerly BabCO) mAb HA.11. There was no staining. Either the preparation of the tissue was bad, or the transgene isn't being expressed. I do think the transgene is expressed because there is a very definite phenotype. Has anyone used FFPE tissue?
