We use Cleaved caspase 3 from Cell Signaling (Ref:9661) and recently we have changed the protocol.
Now we use Buffer EDTA 0.01M (instead of EDTA 1mM) Ph:8 (Microwave: 5 min 800W +15 min 600W cool 20 minutes) in mouse Thymus and we obtain positive staining (overnigth incubation 1:50).
In the previus protocol we used citrate buffer but it doesn,t work always...
We use PBS-T + 5% normal goat serum as antibody diluent
We also have problems with cleaved caspase 9, we are trying now with it with no results,any suggestion for cleaved caspase-9?
