IHC and paraffin embedded C6-GBM cell lines
« on: March 23, 2010, 12:10:05 PM »
Hi,
I have stained formaldehyde fixed parafinn embedded sections of cell lines C6 and GBM for acetyl histone3. I just got staining in a few cells just in the boarders of section. I didn't get any staining in any other cells. Is it an artifact? If so, what causes such an artifact? Is there any better method for IHC staining of cell lines with less artifact and more reliability?

Staining only occuring at the edges of the sections is consistent with drying artifact. The section was not kept in a humid environment for one or more steps of the procedure.

Thank you for your response. I used wet facial tissues to make a wet environment in the mentioned case. From now on I use a wet chamber.
