Staining brain tissue that was never frozen

Hi,

I received human brain tissue, which was perfused with PFA and kept in it for several years. It was never frozen. I wonder what the best sectioning procedure would be.

Can I still cryoprotect the tissue in 20% and 30% sucrose and do cryostat sectioning or should I use a vibratome?

Does too long postfixation impair cryoprotection?

processing, embedding and sectioning procedures should no difference. The brain tissue should work fine for routine histology and special staining. But it may not work well for immunohistochemistry.

It depends on your following testing, but why not embed in paraffin? If you want to stain the brain with procedures, that also work on ffpe-tissue, I would prefer the better morphology of a paraffin-section.
