Bubbles/Condensation
« on: June 01, 2010, 12:10:58 PM »
Hi All
I am having trouble with basically any staining i am doing.
When you look at the slides there are small bubbles (Microscopic), it also looks like there is condensation on the tissue.
I cant attach the pic as its too big!
I am rehydrating to granded alcohols then through a xylene substitute, i mount with DPX and leave overnight to dry!

Any help would be great!

Do you dilute the DPX with xylene? You may have water in the DPX. Coverslip a blank slide and take a look. If you see droplets, dump the DPX, clean your dispensing bottle, mix up fresh mounting media.

Thanks for the quick reply
I dont dilute DPX with xylene, should i?
I usually just purchase a bottle from Sigma  and leave it in it? Is this bad practice?

Are you the only person using your set-up? If not check to make sure that everyone else is also following the correct times and sequence for the dehydrating, clearing and mounting of the slides. I have had some experiences in the past were issues like this suddenly disappeared when a specific person went on holiday.
