tissue detachment

I am having a problem with needle core biopsy tissue comming off the slides. I am currently using super frost plus slides with a little gelatin in the water, the paraffin is sections at 3 microns then air dried standing up and left on a slide warmer flat over night, they are then placed in a 55 degree oven for 1 hour before staining. The sections are stained on an autostainer so manipulation of the tissue should be at a minimum. Does anyone have any idea's as to why some of the tissue is detaching? The slides are a new batch so the charge should be good.

Try skipping the slide warmer and leave them in the oven overnight instead.

How many xylenes to deparaffinize? How long in each? Are they and the alcohols clean? Especially the last ones.

I found putting enough gelatin (i.e. >=0.03% concentration) in water bath will help tissue sections stay on slides. But the potential drawback is higher background staining may occur.

maybe the slides are too good. we use common APES or polylysine coated slides, work well, no detachment. but, super frost slides are superior for frozen section.

Do you notice trapped water under the paraffin slides after the first drying?
Are the superfrost plus slides beyond shelf life? Are they stored open in warm environment?
Would it work better without gelatine?
