Hello everybody!

I am looking for a protocol to stain formalin-fixated, paraffin-embedded human liver samples with ceruloplasmin AB.So far, I just heard about cryosections treated with this AB. Because I do not have much material to practise I am very interested in any ideas how to proceed.

Where did you purchase the antibody from? Is the datasheet telling you anything about the pretreatment or application or suggested dilution range?

I did not buy an antibody yet because I first wanted to find out more about  this antibody and paraffin sections.

Samira,

Based on Tony's posting, this antibody seemed working well on FFPE tissues. You may want to find original papers for a quick reference.

I have the following suggestions regarding the testing of the antibody:

1) Try three pretreatments: Citrate buffer pH6.0, Tris-EDTA pH9.0 and Trypsin digestion.

2) Use diution recommended by manufacture or set a serial dilutions of primary antibody (i.e. 1:100, 1:500, 1:1000).

3) You should have pretty good chance to get this antibody work.
