NeuN antibody neuronal staining

Hi,

I'm planning to use NeuN antibody from mouse to stain rat brain tissue. I want to demonstrate neuronal degeneration of brain after excitotoxic lesions (ibotenic lesions, 6-OHDA etc). Lesions are typically 3 week old.

1. question: is NeuN a better choice than Nissl-Staining? Is it more selective?

2. question: are there special things to take care of with this stain, like fixation etc (I'm planning to do free-floating cryosections on PFA-perfused and shortly postfixated brains)

3. is there a better (= more specific) stain available which you would recommend

no I have not yet tried this myself. Some coworkers tried it out two years ago, and hat lots of problems with it - but this was before my time, so I do not know what went wrong with it.

Do you use Fluoro Jade on a regular basis?

Any hints would be welcome.
