Title: Bouin fixation and TUNEL

Hello! I have to do a TUNEL stain in rat testicle( with ROCHE TUNEL KIT AP). They have to be fixed in Bouin for morphological reasons( in the H&E staining), but i'm wondering if there won't be too much background in my TUNEL-stain, since Bouin is an Acid- containing fixation. Does anyone have experience with Bouin fixation and TUNEL? Does it work perfectly? Or should i rather go for Formaldehyde-fixation and take a small specimen of testicle from the ,,Bouin- portion''. Usually i have good results with Formaldehyd- fixation. Thanks in advance.

I have no personal experience about TUNEL on Bouin fixed tissue. You may find the following paper helpful. In this paper, five different fixatives (including Bouin) were compared for TUNEL staining.
