Title: markers of apoptosis

Hello, I am working  on apoptosis on paraffin-embedded mouse tissues. I used TUNEL and cleaved caspase 3 for the markers of apoptotic cells. Both of methods are working but the spread pattern is different. Therefore my questions are 1) what is different in these two markers? 2) Which one is truly reflect apototic cells? 3) Does cleaved caspase3 located on both nuclear and cytoplasm? 4) what is difference between caspase 3 and activate caspase 3? Thank you very much.
