Title: myosin heavy chain antigen retrieval and ALD-58

I am using ALD-58 in IHC to detect slow-tonic skeletal muscle fibers in sections of amphibian muscles.  Is it possible to do this with formalin-fixed, alcohol preserved museum specimens using retrieval techniques?  Any feedback would be great because I am fairly new to IHC and its uses.  Thanks!

All you can do is try, but they may have been fixed for too long a period of time.

Hi,

I have found that using a deparaffinization of 30 mins x 3 (or more!) in xylene substitute can really make a difference in exposing antigen sites.  That is the technique I use, in addition to varying types of antigen retrieval, when tissues have been in formalin for a long time.

Have you tried the double antigen retrieval?
