Title: Basic TEM questions

I am about to start TEM project and currently learning this technique. A few questions to ask.

1. How big the block should be appropriate size?
2. Osmium for 1 hour. Can I fix overnight?
3. After embedding in capsal, how long should the block incubated in 65 C oven, 24 hour or 48 hour?
4. Do I have to do thick sections before thin sections?

Any help is appreciated.

1. The size of blocks should be around 1 cubic mm
2. No. You can only fix tissue with osmium for 1-2 hours long and no longer.
3. Baking of embedded blocks should be at least 24 hours. I have used 72 hours incubation time without any noticeable problem.
4. You don't need to do thick section if the area of precision is not important to your study.
