I am using tissue that was fixed for 4 hours in 4% PFA and then eventually embedded in OCT and frozen.  Does an antigen retrieval step need to be done?

The vast majority of Antigen Retrieval are done on formalin-fixed, paraffin-embedded tissue sections. Fresh frozen sections are not needed for antigen retrieval. However, some (very few) PFA fixed frozen tissue sections are needed for antigen retrieval depending on the antibodies used. I suggest do a test stain without AR. If it worked, that's it.
