Title: Preabsorption in western blotting

I have been working with membrane protein obtained from brain tissues.  I need to validate a specificity of an antibody.  Antibody gives a single strong band with all tissues.  i would like to do preabsorption with the same.  I have used two preabsorption ratios, 5ug of ab to 125ug of ag and 10ug of ab to 125ug of ag.  I could block the band with 10ug of ab.  Does this sound right and logical?
