Has anyone successfully used sPLA2 antibody in paraffin-embedded tissues (especially in brain)?  I have followed several published protocols and have had no success, even after tweaking with different pH antigen retrieval methods, length of primary antibody incubation (1 hr to over night at both room temperature or refrigerated) and using both the DAB (DAKO) and Envision (DAKO) systems for linking and staining.  I also tend to have a lot of background staining making it difficult to actually visualize cells if they were positive.  The published materials and methods are not very detailed at times, and attempts to contact the corresponding authors for additional information has been fruitless (especially since the articles were published 5 -6 years ago).
