effect of different fixation

For ICC staining (ABC method and DAB kit) cytospins, using 4% PFA & Triton-X 100 or cold acetone fixation should give the same results. Am i right?
Because PFA could cause autofluorescence, so PFA or acetone fixation in IF staining could produce different results. Am I right?

Apple, this is the same question you have already posted a bunch of times!!!  My head is spinning!

Comparing fixation with 4% PFA and TX-100 and aceton eis like comparing apples to oranges no matter what the detection!! You should NOT EXPECT the same result. It may be the same, but maybe not, depending on antigen etc etc.

PFA fixation of cytospins may cause autofluorescence but it should not be toooooo serious. It gets more serious when one is using paraffin sections.
