Count of cells in Neubauer chamber

In my work we are searching isolated tumor cells in bone marrow aspirates. We use Ficoll-hypaque and find a pellet with mononucleocytes. After this we ressupend the pellet in PBS. This time we need to count the cells in the solution before procede the cytospins. We need to have about 1.000.000 cells in 100 microliters...So, my question:
1. Do you know how to calculate the cells using Neubauer Chamber? Do you know some paper that review this issue (Counting cells with Neubauer chamber). I need the exact calcules...
2. Do you this I can do this with automatic counter (the same used to count the WBC in clinical laboratory)?
