70 % ethanol after 4%PFA
« on: May 09, 2006, 11:16:20 AM »
A newbie question, I fixed a brain in 4% PFA 18 hs, but now I do not have time to process it, may I keep it in 70% ethanol for a couple of weeks? if that is possible,  after that all I have to do is cryoprotect it and use it?
thank you very much.

You can store in 70% for even longer. BUT, before you cryoprotect you will need to rinse the alcohol out. It will not freeze at the temp for cutting.

ok, thank you. I was thinking to leave the brain in PB during the day and change it several times to extract all the ethanol.

I would not recommend you store it in ethanol unless you are planning on paraffin embedding. Freezing/processing takes just a few minutes so I strongly recommend you proceed with your protocol if you care about your tissue. But if you really cannot cryoprotect and freeze for a few weeks I suggest that you leave it in sucrose/PBS, just add a tinge of azide to prevent growth of bacteria and fungus.
