Substance P and IHC
« on: March 21, 2003, 09:06:18 PM »
Does anyone know if the Polyclone antibody of Substance P (SantaCruz) works on the parafin embeded slides for IHC ?Most paper reported on cryostat slides, only quite few reported on paraffin slides. In addition, will the short time of decalcification (30min)affect the outcome of the IHC staining?Thank you very much!!!!!

Substance P for Paraffin Sections
« Reply #1 on: March 22, 2003, 12:48:02 AM »
Substance P (N-18) from Santa Cruz (Cat#sc-9758) has not been tested on paraffin sections. But the monoclonal rat anti-Substance P (Cat#sc-21715) may work with paraffin sections.

Substance P antibody (ab6338) [NC1/34 HL] from Abcam works for formalin fixed, paraffin embedded tissue sections. You may need to use enzymatic antigen retrieval method to uncover the antigen.

Another source is Chemicon (Substance P antibody, Cat# AB1566). This antibody also works with formalin fixed, paraffin embedded tissue sections.

You may also want to check this article for a reference: http://www.ncbi.nlm.nih.gov/entrez/query.fcgi?cmd=Retrieve&db=PubMed&list_uids=6084658&dopt=Abstract

Decal for a short time will not affect much for antibody binding. But you may need to do a test for different timing.

Hope this helps.
