Need Help finding a general counter-stain for whole cell
« on: January 28, 2008, 11:59:54 PM »
Hello everyone,

This is my first post to these boards. I have a specific question and any help would be greatly appreciated!

I have plated some human astrocytic type cells onto glass and fixed them in 4% para and I have stained them using Alexa488 (ab 495 em 520) and Cy3 (ab 550 em 570).

Now I am need a general cell stain that will stain the entire cell but one that will not interfere with the two above mentioned fluorophores.

This cold in theory be any dye - such as one that could be seen under light-microscopy or even a dye that only fluoresces in UV. The main requirements are that it is the entire cell and it does not interfere too much with the above mentioned fluorophores.

Any advice would be greatly appreciated!

Many Thanks,
Michael

Counterstain normally stains nuclei of cells. It should be good enough for overall morphological demo of the tissue/cell's basic structure.

DAPI may be suitable counterstaining dye for Alexa 488 and Cy3.

cellomics whole cell stains will do the trick.
