Secondary anti-rabbit
« on: July 17, 2008, 08:13:52 AM »
Hi,

I need to buy a biotinylated secondary anti-rabbit. Any suggestions on what is the best antibody with the lower background. I used befor a goat anti-rabbit from Vector but it was too much background.

I use goat anti-rabbit from DAKO (http://www.dako.com.au/prod_downloadpackageinsert.pdf?objectid=108477001). It works quite well for my mouse tissue.

Try using Dako's Envision or Vector's ImmPress (less expensive) detection. They do away with the secondary step and there is no avidin/biotin background.

I have used Vectors ImPress before and got some strange background with some antibodies compared to the classical ABC method. Otherwise it would be very convenient and time/sparing

The best most specific antibodies on the market with the widest range of applications are from Jackson ImmunoResearch. They sell anti-rabbit IgG secondaries which are highly cross-adsorbed against many species. There are of course good LSAB kits from DAKO and other similar places but they may or may not be suitable for a wide range of species (not sure if that matters to you).
