Luxol Fast Blue on unfixed tissue sections

(1/1)

irenes:
Does anyone knows if I can do Luxol fast blue staining on unfixed frozen brain sections?
What do I have to do different from a normal protocol for frozen sections?
Thank you in advance.

richard03:
I do not think you can do Luxol fast blue staining on unfixed frozen brain sections. Why do you need to do unfixed?

CanuckPhD:
According to Kiernan "Histological and Histochemical Methods" (great book IMHO) you can place unfixed frozen sections in 100% ethanol prior to placing in Luxol solution (which is in 95% EtOh). We have done this in the past and it works okay but the results are not as good as FFPE. Be careful with the differentiation as it is often times quicker then with FFPE.

You can also try a solochrome stain with a methanol fixation prior to the staining. I think it is a better and much quicker stain then LFB.
