Hi,

Does anybody know any protocol how to perfuse rats with 4% paraformaldehyde to get their brains fixated and run stereology?
How much PBS and paraformaldehyde, temerature etc...?

We perfuse with 50 mls of PBS and then 100 mls of 4% PFA, remove brains and then postfix for another 24 hours. This gives us good results. However the perfusion speed is critical as if you go to fast you can damage the vasculature. With our perfusion pump we perfuse at 8 mls/min. However, you must check your pump and make sure that the force is not too great at this speed, due to tubing size.
