Western blot IHC problem

Hello all,
   I am woking on a protein which shows higher levels of expression in tumor tissue than in normal tissue on western blots.   When I did IHC on For. Fixed-Par. Embedded Normal and Tumor tissue, they both stained positive.  I lowered the concentration of 1' Ab, but I lose the signal in the tumor tissue first.  Can this be right?

Jason,

What proteins you are working with? Some studies showed that some epitopes in the tumor tissue were somewhat more susceptible than that of the normal tissue to decalcification, formalin fixation and paraffin embedding. That may be an explaination of why you lost staining in tumor tissue first.
