paraffin sections
« on: September 27, 2010, 12:52:08 PM »
Hi all

I am usually dealing with frozen sections and I had to deal with some paraffin sections  these days, I have been told that there is a step for antigen retrieval which is the only difference between both types of sections other wise every thing is  the same in terms of the staining procedures. Is this true?  and what is the protocol for antigen retrieval ?

many thanks for your help in advance

I would recommend the DAKO Handbook. You can find it online under literature on the dako-website.
And you can find retrieval procedures on this website under protocols.

And not all antibodies done on paraffin sections need antigen retrieval. The datasheet should state what is recommended.
