So I have never done this type of immuno before.  We want to stain cells and blood vessels.  Do I need to fix these?  I have worked strictly with fixed tissue in the past (PFA/Sucrose).

Please advise the best way to preserve these and follow through with Immuno.

Fixation normally depends on your antibody. You can find out what fixation to use on the antibody datasheet. Normally they have a protocol section. IF they don't then call the company for more information.
