Sodium borohydride
« on: October 23, 2003, 02:47:11 PM »
Hi

What is optimum and safe concentration of sodium borohydride to use (and incubation time) for breaking aldehyde linkages and get good immunostaining?
Does the concentration of 1% borohydride is highly drastic for antigens compared to  0.1%?

Thanks

I always use 1% sodium borohydride in 0.1M PB, and incubate sections for 30 min at room temp. Then wash 3-6 times or until bubble is gone.

I use 1% sodium borohydride in water and incubate sections two times,  for 5 minutes and again for 15 minutes. then wash 3 times un PBS.
