how to clean OCT from cryosection completely?

I have trouble with removing OCT from slide when I wash it by water or PBS. If anybody have any idea what's wrong may be responsible for this little trouble? I did regular step by 2% para fixation and 30% sucrose and OCT embedded, stored at -80C, cut in cryostat...dry in room temp for at least 5 h...

OCT is water soluable so it should be removed easily by wash in water or PBS. From the protocol you provided, the drying time (5 hrs) seems too long (normally half a hour will be sufficient). Also 30% sucrose is really NOT necessary for OCT embedded tissues. So that may be a potential factor as well. Otherwise, the OCT you purchased is JUST a bad batch.

Thanks, Susan. I used 5h dry, because my tissue was very easy to be washed off with less dry hour. For 30% sucrose, the slides will be stored at -80 degree. I tried again, OCT is still sticky around tissue. The problem is the same tissue I did before was always good, but recently bad, I'm wondering what's wrong with my procedure.
