hi, I did frozen section staining for 1 year. Recently, the tissue is very easy to fall off from the slide (Gelatin coated) after 37C 1h incubation. I did all same as before, cut into 10um section, air dry and stored in -80C. I give it half to 1 h for warm after taken from freezer, then put it into PBS, proteinase K 10min, 3%H2O2 5min, normal serum 30min, 37C 1h...

Any wrong step or any reason is possible?

The proteinase K digestion step may contribute to sections coming off slides. Is this necessary step? You may want to reduce proteinase K concentration or using lower incubation temperature.
