What replacement for DAB (Xylene mounting)?
« on: August 26, 2004, 09:38:30 AM »
Hi

I am looking for a replacement for cancerogenous DAB, without having to go for aqueous solution.

That means I want to dehydrate and mount my slides as I would with DAB: alcohol / Xylene.

Any suggestions?

Would the Hanker- Yates peroxidase reaction be a solution to this problem?

Is anyone using it?

Thx,

Dierk

I don't know about replacing the DAB, but if you are looking to get rid of all your HAZ MATs, you can find a product called Histoclear which does the same thing as Xylene with out any of the regulatory problems (ie. non flammable, non hazardous).

You can use AEC, rinse in water, then place a few drops of Crystal Mount on them, let them dry, then dip in xylene and coverslip with permount. I even use Crystal Mount on fat stains done on frozen sections with no lose of staining.
