activated Caspase antibodies

Has anyone used these succesfully or otherwise?  I am having problems geting polyclonal activated caspase 3 and 9 to work. also would be greatful of suggestions for antibodies I can use as controls for these in parafin sections in microwave, pronase and no pre treatment antigen retreaval controls

Proteolytic pretreatment can be very tricky in my experience. Have you tried HIER in Alkaline buffer like TRIS-EDTA?

Cleaved caspase 3 from Cell Signaling works very well in steamer HIER with 1mM EDTA, pH 8.0  as the retrieval solution.

Rb anti-Activated Caspase 3

Hi,
I am using B/D & BioVision polyclonal ab; Rb anti-Activated Caspase 3 in frozen mouse eyes, and they work great with 1:50-1:100 dilution with 2 steps Flurorescence IHC, but incubation overnight @ room temperature instead 4C.

We use Cleaved caspase 3 from Cell Signaling (Ref:9661) and recently we have changed the protocol.
Now we use Buffer EDTA 0.01M (instead of EDTA 1mM) Ph:8 (Microwave: 5 min 800W +15 min 600W cool 20 minutes) in mouse Thymus and we obtain positive staining (overnigth incubation 1:50).
In the previus protocol we used citrate buffer but it doesn,t work always...
We use  PBS-T + 5% normal goat serum as antibody diluent
We also have problems with cleaved caspase 9, we are trying now with it with no results,any suggestion for cleaved caspase-9?

We use the Cell Signaling #9661 antibody. It works great on injured mouse tissue.
