Hematoxylin in hippocampus
« on: May 25, 2012, 11:48:20 AM »
Hello. We are doing ABC-DAB stains of phosphorylated ERK in the hippocampus. We would like to counter-stain with hematoxylin to verify neuronal nuclei. From what I understand, it should be applied once the sections have been mounted but prior to dehydration. So, is the idea to plate the pERK sections, let them air dry, apply the hematoxylin, rinse, and then immediately dehydrate and cover-slip? Any clarification would be appreciated. Thanks.

After chromogen development you rinse the slides in dest. water, then put them into the hematoxylin solution, followed by bluing in tapwater.
Then dehydration through graded ethanols, then clearing in xylen (or similar) and coverslipping.
The hematoxylin solution may be diluted, or duration shortened to give a light nuclear counterstain.
