Tracking cell migration/proliferation by IHC/IF?
« on: February 19, 2014, 01:27:16 AM »
So my question concerns tracking cytoplasmically labeled cells adoptively transferred to mice for cell proliferation assessment. This is usually done with flow cytometry on single cell suspensions, but I would like to take some tissue samples for histologic confirmation of disease and also to visualize the tissue localization of the infiltrating cells. I will be using CellTrace Violet as my cell tracer. Can this survive formalin fixation and paraffin embedding? What about PFA and cryosections? I cannot use CFSE+anti-fluorescein Ab because GFP is also being used in these experiments. Additionally, any tips on the best nuclear dye (preferrably red) given that CTV will take the place of Hoechst/DAPI. I know about DRAQ5 but I was wondering about any other dyes that have given good results.

Any help much appreciated.
