B-Gal and paraffin processing.
« on: September 16, 2014, 06:48:32 AM »
Hi
I have a researcher who is looking at B-Gal staining in paraffin tissues. The problem is that xylene removes the B-Gal. There is some literature that says you can substitute xylene with normal Butanol. However I cannot find a protocol . Can anyone help?
Many Thanks
J.

I am not familiar with this special stain, but you can try to mount the section with aequaous media like "Aquatex" from Merck. So you don't have to go through ethanols and xylene, but I don't know if the staining product is stable with this method.
