Complete Eosin removal before IF


Hi there,

I was wondering whether anybody around here knows a method for the complete removal of Eosin from FFPE sections - the method has to be compatible with IF.

I get my samples from an institute of pathology that routinely stains their tissue samples with Eosin (so you do better see them if they are small). However, Eosin is fluorescent in FITC as well as TRITC channel, which means I have a very strong background, as I work with human colon samples - which contain many eosinophils.

I tried incubating in water or EtOH where Eosin is soluble - however it seems that only the unbound Eosin is removed.

I would really appreciate it if somebody could help me:)
