H&E counterstain for Oil Red O
« on: July 24, 2015, 02:03:51 PM »
I have a client who wants to stain frozen sections with Oil Red O, and he wants to use H&E as a counterstain.  Does anyone have a protocol for this?  Thanks.

I have no special protocol for you. But have in mind, that ethanols and organic solvents solve the stained fat. So you have to avoid those. With eosin there is a special problem, because it bleeds out in water-based mounting medias and acryl-based medias cannot be used because of the solvage of fats.

If I were you I would talk to your client about the probably non-optimal results before trying.

If you only want to do a nuclear stain with hematoxylin. Just 5 min in Mayers hematoxylin, bluing in tapwater and mounting with water-based media.
