I'm new to this forum and also quite new to the world of research, so I hope this won't be too stupid to ask...
I have DAB stained coronal brain sections and would like to compare the number of neurons in the cortical layers between two groups. I have eight sections from the same bregma for each individual.
My idea is to define ROIs that have a fixed width and have variable length that covers all cortical layers. I would randomly place a ROI in the somatosensory cortex and count the neurons manually for each layer. Then I would like to compare these cell counts.
Is this an acceptable way to do it and is it reasonable to conclude whether or not there is a difference in the amount of neurons between the two groups?
