I'm trying to do an Immunohistochemistry on paraffin embedded mouse spleen (used as positive ctrl because of erythrocyte presence) and on mouse peripheral nervous tissues. I obtained an unexpected result: a poor staining of erythrocytes (which must express SLC4A1 on membrane) and a diffuse staining of cell nuclei of different tissue cells (splenocytes, neurons, etc).
Being SLC4A1 a trans-membrane protein this is a very strange localization...Have anyone ever observed this kind of nuclear localization?
Alternatively, do you know a good Ab for immunolabeling SLC4A1 on mouse paraffin embedded tissues?I'm working with this Ab: rabbit anti-band 3 18566-1-AP Proteintech.
