Hello everyone !

By misstake I made a combined Antigen Retrieval , first microwave boiling in citratbuffer pH6.0 3x5 min after that 5 min in trypsinsolution 0,1 % 37deg.

I discover that I had some staining in these slides but not in the ones that only had microwave or trypsin only.

Does anyone have experience in combining these two ?

Sinse it was an antibody I haven't used before I'm not sure if the staining is true or false .

Combining of antigen retrieval methods has been studied and practiced recently and it is a good approach for IHC staining of certain antigens that can not be recovered using single HIER or PIER.

I have had a very good success with Collagen IV staining using HIER and PIER combined method. The example of protocols is listed below:
