Hello one and all,
I would like to do some fl immuno with MAP2 (1:1000) and Alexa Fluor 488.  Th other label I am using is GFAP with CY3.  The protocol I have uses 0.3% Triton X and 10 % donkey serum.  However it did not work.  Should I stain for the GFAP first then MAP2 or is there a better detergent to use for this?\

Any suggestions would be greatly appreciated.  It seems like I am always trying to trouble shoot my immuno.  Is this normal?

The order is not as critical as the concentration and method used. Did the antibody work on single labeling? If not, you should try to work for the protocol and method first for each antibody separately before the double labeling. GFAP should be easy to work with.
