PROBLEM NUMBER 29

Poor staining or lack of staining with alcian blue dissolved in acetic acid and commonly used for staining mucin.

 

WHY?

 

Alcian blue is said to denature in acid solution over a period of time, the stronger the acid component the sooner denaturation occurs. For example it is said that in a 3% acetic acid solution alcian blue denatures in approximately 1 month.

 

So the answer is to make up the solution more frequently.
